similar yield as the standard method. Therefore, using this modified method, we can reduce the soaking time from 15 h to about 10 min. In summary, a simple modification of the crush-and-soak method allows the recovery of ODN from polyacrylamide gel within 30 min with high yield. The key difference in our procedure is the replacement of the overnight soak with a heat/freeze step (summarized in Table 2). We assume that heating causes the crushed gel to swell rapidly, hastening the diffusion of the ODN. Subsequent freezing may shrink the solid gel matrix while expanding the water volume, thus squeezing the ODN out of the gel network. This modified method is simple and fast, yet produces similar recovery as the standard overnight soaking method. It provides a rapid alternative to the standard crushand-soak method for recovering either labeled or unlabeled oligonucleotides from polyacrylamide gel.

Sequence determination of the capsid protein gene and flanking regions of tobacco etch virus: Evidence for synthesis and processing of a polyprotein in potyvirus genome expression.

The nucleotide sequence of the 3'-terminal portion of the tobacco etch virus (TEV) genome was determined. The 2324-nucleotide sequence represented approximately one-fourth of the TEV genome and included the capsid protein gene and flanking regions. An open reading frame of 2135 nucleotides and an untranslated region of 189 nucleotides adjacent to a polyadenylate tract were identified. The seque...

Characterization of the rubella virus nonstructural protease domain and its cleavage site.

The region of the rubella virus nonstructural open reading frame that contains the papain-like cysteine protease domain and its cleavage site was expressed with a Sindbis virus vector. Cys-1151 has previously been shown to be required for the activity of the protease (L. D. Marr, C.-Y. Wang, and T. K Frey, Virology 198:586-592, 1994). Here we show that His-1272 is also necessary for protease ac...

Penicillin-binding protein 1B from Escherichia coli contains a membrane association site in addition to its transmembrane anchor.

A working structural model of penicillin-binding protein 1B (PBP 1B) from Escherichia coli derived from previous data consists of a highly charged aminoterminal cytoplasmic tail, a 23-amino-acid hydrophobic transmembrane anchor, and a 758-amino-acid periplasmic domain. Using an engineered thrombin cleavage site, we have investigated the solubility properties of the periplasmic domain of PBP 1B....

Sequence determination of the Sendai virus fusion protein gene.

From a genomic DNA library of Sendai virus, we have identified and sequenced clones corresponding to the F glycoprotein gene. The limits of the F gene region were defined by mapping the 5' and 3' ends of the mRNA with S1 nuclease. The Sendai virus F gene is 1821 nucleotides long. The predicted primary translation product of the single long open reading frame would code for a protein of 565 amin...

Purification and Characterization of the Mouse Mammary Tumor Virus Protease Expressed in Escherichia coZi

The mouse mammary tumor virus (MMTV) protease gene was cloned into pGEX-QT, an Escherichia coli expression vector containing the glutathione S-transferase coding region of Schistosoma japonicum. The chimeric protein was formed by fusion of the glutathione S-transferase with a hexapeptide which contains a thrombin cleavage site, followed by the MMTV protease. Affinity chromatography on a glutath...
